Definition: What is ALCOA+?

ALCOA+ is a framework consisting of nine principles for data integrity in GMP-regulated environments, recognized by the FDA and the EMA. It requires that data—whether recorded on paper or electronically—be attributable, legible, timely, true to the original, accurate, complete, consistent, persistent, and available—and forms the substantive foundation upon which technical requirements such as EU GMP Annex 11 or FDA 21 CFR Part 11 are based.

Who is ALCOA+ intended for?

These principles apply to all companies that generate, process, or store data as part of GMP-regulated processes—in particular, pharmaceutical manufacturers, active ingredient manufacturers, and contract research organizations. They can be applied regardless of production methods or systems, ranging from handwritten batch records to fully digitized manufacturing processes.

  • Original (true to the original): primary data or a verified copy; no unauthorized intermediate steps

  • Accurate: Values that arecorrect , error-free, and consistent with the actual event

  • Complete, Consistent, Enduring, Available: records that are complete, internally consistent, permanently preserved, and accessible at any time

Difference from EU GMP Annex 11

ALCOA+ describes the fundamental, system-independent principles of data integrity. It defines the characteristics that data must have, regardless of how it is generated. EU GMP Annex 11, on the other hand, specifies how computer-based systems in particular must technically implement these principles: through validation, audit trails, access controls, and electronic signatures. Put simply, ALCOA+ is the substantive goal, while Annex 11 is a technical means of achieving that goal using digital systems.

Consequences of Noncompliance

Violations of ALCOA+ principles are among the most common and serious types of findings during GMP inspections by the FDA, EMA, and national authorities—such as handwritten corrections without a countersignature, values added retroactively, or incomplete batch records. Such data integrity deficiencies are considered particularly critical because they raise fundamental doubts about the reliability of all production data and can lead to batch holds, warning letters, or the revocation of manufacturing authorization.
