Definition: What is FDA cGMP?

FDA cGMP (Current Good Manufacturing Practice) refers to the U.S. requirements for processes, equipment, personnel, and documentation in pharmaceutical manufacturing, as regulated in 21 CFR Parts 210 and 211. Unlike specific individual regulations such as Part 11, cGMP covers the entire production system: from raw material testing through manufacturing processes and cleaning validation to final inspection and batch release.

Who is subject to FDA cGMP?

cGMP applies to all manufacturers of drugs marketed in the U.S.—regardless of whether production takes place in the U.S. or abroad. This applies to both manufacturers of finished drug products and suppliers of active pharmaceutical ingredients (APIs) and raw materials that fall within the scope of the regulations as part of the supply chain.

Difference from EU GMP

FDA cGMP and EU GMP share the same fundamental objective—consistent, quality-assured drug manufacturing—and are largely identical in content, as both are based on the same international principles (including ICH guidelines). The formal difference lies in the legal basis: cGMP is U.S. law codified in 21 CFR Parts 210/211, while EU GMP is regulated in EudraLex Volume 4 and is directly applicable in the member states. Through mutual recognition agreements such as the EU-U.S. Mutual Recognition Agreement, both regions are increasingly recognizing each other’s GMP inspection results, which reduces the double burden on international manufacturers without harmonizing the regulatory frameworks themselves.

Consequences of Noncompliance

cGMP violations are among the most common causes of FDA warning letters and Form 483s. In the event of serious or repeated deficiencies, the FDA may prohibit the import of affected products into the U.S. (Import Alert), suspend market authorization, or, in extreme cases, initiate criminal proceedings. Since cGMP inspections can occur without prior notice, it is essential to maintain a state of readiness for inspections at all times, rather than only when prompted by specific events.
